1. Use current source ( 0.1mA ) to charge a capacitor.
2. After disconnecting the current source, measuring the flowing current from that capacitor.
I want to use an analog buffer to deal with the potential overshoot, but the requirement of this transmission line is that the leaking current produced by the analog buffer can not be higher than 0.001mA.

Almost any run-of-the-mill Op Amp can do this, as long as it's used in the non-inverting configuration. The leakage current will be, at most, the Op Amp bias current plus the Op Amp p0ffset current.
