What does it mean by Gain & Offset ? By rotating the screw of pot. of Gain & offset, What would happen ?

Can you post the circuit/example you are referring to?

In general "gain" would refer to multiplication of a signal and "offset" would refer to addition to a signal, i.e. out = in * gain + offset, but it is not possible to be sure that is what you are referring to without seeing context.

Remember the simple mathematical equation of a straight line

y=mx+c

m is the gain and c is the offset.

In case of an audio amplifier circuit, m decides the amplification factor i.e. the volume control knob controls the gain. c decides that when the gain is zero, i.e. when the volume control knob is at zero, the amplifier still produces audible sound if offset is greater than zero.

In the case you are refering to, the gain is the signal amplification and offset is the signal at the output when gain is zero.
