In the development of DAQ system is necessary a calibration?

I am planning to build a DAQ system to make some voltage measurement in the lab. My aim is to measure the Vth of a Mosfet.
I am developing a DAQ system made by an FPGA and an ADC. Is it necessary to calibrate the system? Can I trust just the datasheet? .
I want to put an instrumentation amplifier at the input but it is not necessary. Now the analog inputs are straight in the ADC.

You can believe the datasheet, but can you believe your design? You most certainly need to do some testing to verify that your circuit is working; why wouldn't you? Are you proposing just throwing some components on a PCB and hope that they are all connected properly?

You make no mention of your required accuracy and resolution.
