Can anyone tell me how to estimate the FFE once I have recorded the guitar sound in MatLab?

How long should I record for a note?
Is it enough if I record for five-second?

Search for help on the rootmusic function (Signal Processing Toolbox) to see if it meets your needs.

The longer your note, the better the frequency measurement. Five seconds seems plenty long for most applications.
